Recently, Professor Lv Chulun, School of pharmacy, Nanjing Medical University, as the corresponding author, published a research paper entitled "the 3 'untranslated region protections the heart from angiotensin II induced cardiac dysfunction via aggf1 expression" on the internationally famous journal molecular therapy.
Hypertensive heart disease is a heart disease characterized by abnormal left ventricular function. It has high incidence rate and mortality worldwide. Finding the target drug to protect the heart is the key to clinical treatment. It was found for the first time that 3'UTR has the ability of CIS regulation in the process of cardiac protection, which can regulate the expression of myocardial protective protein, improve cardiac function and play a more protective role in the heart.
In this study, mice with hypertensive heart disease were treated with aggf1 full sequence virus containing 3'UTR sequence. It was found that aggf1 full sequence virus containing 3'UTR has stronger heart protection function than aggf1 (a vascular growth factor with heart protection function) coding sequence virus. Further study of its mechanism showed that there were a large number of microRNAs (microRNAs) expression increased under the pathological / angiotension II perfusion of hypertension, and some of them could recognize the 3'UTR of aggf1 and MCL1, which decreased the expression of aggf1 and MCL1, leading to cardiac dysfunction. Therefore, when the aggf1 full-length sequence virus with 3'UTR was used for intervention, the down-regulation of endogenous MCL1 could be inhibited, so as to improve the heart function. This study provides an important target for the clinical treatment of hypertensive heart disease.
