In March 2020, Professor Ji Yong, vice president of Nanjing Medical University and director of Jiangsu Key Laboratory of cardio cerebrovascular medicine, School of pharmacy, published a research paper entitled "hydrogen sulfide advocates liver intervention via s-sulphydrated-keap1 / Nrf2 / LRP1 pathway" in the form of article on Hepatology, the top international journal. The corresponding author of this research paper is Professor Ji Yong, the co corresponding author is Xie Liping, associate professor of Department of pathophysiology, School of basic medicine, Nanjing Medical University, and the first author is Dr. Zhao Shuang, Key Laboratory of cardiovascular and cerebrovascular medicine, Jiangsu Province.
Liver disease is characterized by high incidence rate and easy repetition. How to delay and treat liver diseases is an urgent problem in clinical practice. Sulfhydryl modification of proteins mediated by hydrogen sulfide (H2S) may be involved. Professor Ji Yong's team found for the first time that exogenous administration of H2S donor (gyy4137) can improve liver injury caused by different factors by sulfhydryl modification of Keap1. Hyperglycemia, hyperlipidemia, and drug stimulation (carbon tetrachloride, CCl4) can lead to liver damage, accompanied by a decrease in the level of H2S in the body. Exogenous H2S supplementation can modify the 151 cysteine site of Keap1 (cys151) by sulfhydryl sulfurization, promote the dissociation of Keap1 and Nrf2, and then activate the Nrf2 signal pathway. On the one hand, the activated Nrf2 can promote the downstream antioxidant stress protein (HO-1, GCLC, On the other hand, it can promote the expression of LDLR-related protein-1 (LRP1), alleviate the lipid deposition in the liver, and thus play the role of liver protection. This study provides more possibilities for the clinical treatment of liver diseases.
